John married Christian ERSKINE, daughter of Robert ERSKINE 4th Lord Erskine, 16th Earl of Mar and Isobel CAMPBELL, 25 January 1535(contract).1 (Christian ERSKINE was born about 1507 and died after 20 May 1564 1.)

  Marriage Notes:

"This laird of Luss had been married only about two years previous to his accession to the family estates on the death of his father. The lady whom he espoused was Christian Erskine, daughter of Robert Lord Erskine, who was killed at Flodden in the year 1513, by his wife, Dame Elizabeth Campbell, Lady Erskine. The contract of marriage between John Colquhoun, younger of Luss, and Christian Erskine, is dated 25th January 1535, and the marriage was to be solemnized in the face of the holy kirk within twenty days thereafter. By the contract, Humphrey Colquhoun of Luss became bound to infeft his son John, and Christian Erskine his wife, in as much of the lands of Luss as would yield yearly 140 merks to them in liferent, and to the heirs-male of their marriage ; and Lady Erskine became bound to pay to Humphrey Colquhoun 2000 merks of tocher with her daughter Christian."

from The Chiefs of Colquhoun 1

John next married Agnes BOYD, daughter of Robert BOYD 5th Lord Boyd and Margaret COLQUHOUN, about 3 November 1564.1 2 (Agnes BOYD was born about 1543 and died on 18 July 1584 in Edinburgh, Midlothian, Scotland 1 3.)

  Marriage Notes:

"Sir Matthew married ... second, Jean, eldest daughter of Sir John Colquhoun of Luss, by Agnes Boyd his wife."

from Scots Peerage (vol 2)

"Agnes, married, as second wife, to Sir John Colquhoun of Luss, who can have been a widower only a month or two at most. Being related within the forbidden degrees, a dispensation was granted by John Hamilton, Archbishop of St. Andrews, the Papal Legate, 3 November 1564. He died January 1574-75. She died at Edinburgh 18 July 1584. Her testamentdative and inventar of goods was given up by her son, Sir Humphrey Colquhoun, and confirmed 18 April 1588."

from Scots Peerage (vol 5)

"John Colquhoun of Luss did not remain long a widower. Within a few months apparently after the death of his first wife, Christian Erskine, he married, secondly, Agnes, daughter of Robert fourth Lord Boyd. Sir John and Agnes having been related to each other in the third and fourth degrees of consanguinity, a Papal dispensation was necessary, according to the canons of the Roman Catholic Church, to render their marriage lawful. This dispensation was, therefore, applied for from John Hamilton, Archbishop of St. Andrews, the Pope's legate a latere, who had the power of granting it, and it was readily obtained. This dispensation is dated 3d November 1564."

from The Chiefs of Colquhoun

Note
There is a discrepancy in the above quotations about whose daughter Agnes was. Both the fourth and the fifth Lord Boyd had marriage ties with the Colquhouns, and so it is not difficult to see why a dispensation was necessary.
The fifth Lord Boyd's marriage contract was dated 1535.
